sharing the small set of references I actually re-open when a titration question comes up in the thread. not exhaustive, just the ones that have earned the re-read. on PK and dose-response: the SURPASS-1 through SURPASS-5 primary papers are the obvious starting point, but the more useful read is the phase 1 multiple-ascending-dose work, which is where the half-life (~5 days) and the rationale for weekly dosing actually lives. the registration trials assume you’ve already absorbed that, and a lot of the titration confusion downstream comes from skipping it. on the 4-week step interval: worth reading the SURMOUNT-1 protocol appendix rather than the headline paper. the interval is described as a minimum, and the language matters for anyone treating it as a fixed schedule. on compounded vs branded: the FDA’s 503A guidance docs and the few comparative analytical papers on salt-form variability are dry but load-bearing. the excipient axis is mostly absent from the published literature, which is itself the point. threads here I’ve found useful: the long titration tolerability threads from late winter, and anything where someone’s posted their week-by-week sides honestly rather than retrospectively. happy to add to this if others have references that earned a re-read.
